Linear Free-Energy Relationship of Nonaqueous Biocatalysis - Analysis of Solvent Effects on Enzyme Enantioselectivity
A multiparameter correlation method for predicting the effects of solvents on enzyme enantioselectivity in nonaqueous organic media is presented. Based on our previous kinetic model and the linear solvation energy relationship (LSER), a correlation equation which relates the enzyme enantioselectivity in nonaqueous organic media to dispersion, polarity, acidity, basicity, and Hildebrand solubility parameters of the solvents was derived. Analysis of experimental data reported in the literature showed that the LSER equation is adequate for expressing the dependence of enzyme enantioselectivity on the properties of the solvents. On the basis of the correlation equation, the molecular interactions that influence selectivity in solvents were further examined.
