인슐린 전달용 키토산 팻치에서 피부활성제의 혈당 저감효과
The Effects of Skin Activating Agents on Blood Glucose Level in Transdermal Insulin Delivery System Using Chitosan Patch

키토산으로 만든 약물 팻치제는 조건에 따라 약물방출을 조절할 수 있으며 피부 활성제의 첨가로 약물 방출 능력을 증가시킬 수 있었다. 본 연구에서는 키토산을 약물방출 제어용 고분자막으로 이용하여 in vivo 실험을 하였다. 당뇨병 치료제인 인슐린을 저온에서 키토산을 이용한 팻치로 제조하여 각각의 약물특성과 약물방출 거동을 알아보았으며 나아가서 NOD (Non Obese Diabetic Inbred) mouse를 이용하여 키토산 팻치에 피부활성제의 첨가에 따른 최적의 조건을 찾았다. 키토산-인슐린 약물과 피부활성제(glycerol, DMF, DMSO)를 사용하여 분석한 결과 현저한 혈당의 저감 현상을 볼 수 있었다. 결과적으로 인슐린 팻치에 의한 약물투여는 피하주사 투여와 비교할 때 인슐린의 측면에서 약 8%의 약물 전달 효과를 가져 왔으며, 낮은 혈당치를 효과적으로 유지할 수 있었다.
The drug delivery and release activity from chitosan patches can be controlled and accelerated by skin activating agents. The study was performed by utilizing chitosan as a high molecular membrane, to control drug release in vivo. Characteristics of chitosan-insulin patches and its behaviors in the drug release were examined at low temperature for a possible application in diabetes. NOD (Non Obese Diabetic) mice were used to find optimal condition for chitosan-insulin patches with surface-activating agents. When chitosan-insulin patches were used with the surface-activating agents (glycerol, DMF, DMSO), a considerable decrease in the blood sugar level was observed. The results showed that the patches enhanced 8% increase in insulin over the hypodermic injection; and moreover, the patches can be used to maintain the blood glucose at a low level effectively.
