Engineering a Doxorubicin/daunorubicin Biosynthetic Gene Cluster from Streptomyces Peucetious ATCC 27952 for Development of Hybrid Antibiotics with Improved Resistance
Streptomyces peucetius ATCC 27952 is responsible for producing for producing clinically important chemotherapeutic agents, daunorubicin (DNR) and doxorubicin (DXR), adraimycin. We focused to develop hybrid doxorubicin by modifying the daunosamine sugar with other sugar moieties. The attachment of sugar moieties is facilitated with same glycosyltransferase of S.peucetius ATCC 27952 or without using its own glycosyltransferase. Proper modification on gene cluster of daunosamine by gene(s) from other biosynthetic gene cluster of deoxysugar leads to various hybrid doxorubicin. We expect that introduction of doxorubicin/daunorubicin resistance gene, drrC into wild type S.peucetius ATCC 27952 by pSET152 vector assisted with ermE* promoter enhances the production of antibiotics.
