Enhancement of Biocatalytic Activity of Recombinant Pichia pastoris for the Production of Chiral Epichlorohydrin by Lyoprotectant and Detergent

Rhodotorula glutinis의 epoxide hydrolase가 재조합된 유전자재조합 Pichia pastoris의 동결건조 생촉매 제조과정에서 고활성을 얻기 위하여 다양한 lyoprotectant 및 detergent 첨가 효과를 분석하였다. Tween 20 (5%(v/v)) 등을 첨가한 경우에서 단순 동결건조에 비해 약 2.5배 향상된 가수분해 속도를 얻었으며, 입체선택성도 약 1.9배 향상시킬 수 있었다. 100 mM 인산염 완충용액에서 20 mM의 라세믹 epichlorohydrin에 대하여 약 10 min의 회분식 분할반응을 통해 이론 수율 대비 76%의 높은 수율로 광학순도 100% ee의 (R)-epichlorohydrin을 얻을 수 있었다.
The effect of adding lyoprotectants and detergents on the biocatalytic activity of a recombinant Pichia pastoris harbouring the epoxide hydrolase of Rhodotorula glutinis was investigated. The additionof 5%(v/v) Tween 20 during lyophilization procedure increased the initial hydrolysis rate (2.5-fold) and enantioselectivity (1.9-fold) of the recombinant Pichia. Enantiopure (R)-epichlorohydrin was obtained with a high enantiopurity (100% ee) and a yield of 76% from its 20 mM racemates using the recombinant Pichia.
