생체적합성과 생분해성을 갖는 폴리에테르우레탄우레아의 합성과 물리적 성질에 관한 연구 : (II) 실리콘을 함유하는 폴리우레탄의 in vitro 혈액적합성
Synthesis and Physical Properties of Biocompatible and Biodegradable Polyetherurethaneurea : II. In Vitro Blood Compatibility of Polyetherurethaneurea Containing Polydimethylsiloxane Segments
초록
메틸렌디페닐이소시아네이트 (MDI)와 폴리테트라메틸렌글리콜 (PTMG)로 부터 합성한 폴리우레탄 prepolymer를 에틸렌디아민 또는 양말단에 아미노기를 갖는 폴리디메틸실록산 (ATP-DMS)으로 사슬연장하여 폴리에테르우레탄우레아 (PU) 및 실리콘을 함유하는 폴리에테르우레탄우레아 (PU-S)를 합성하였다. PU와 PU-S의 in vitro 혈액적합성을 조사하기 위해 합성폴리우레탄과 헐액성분과의 상호작용을 조사하였다. PU-S (22-24%)에 점착한 혈소판의 양은 PU (32%)보다 적었다. 또한 점착혈소판으로부터의 세로토닌 방출량은 PU보다도 PU-S에서 더욱 억제되었으며, 이들 결과는 주사형전자현미경에 의한 형태학적 결과와도 잘 일치하였다 한편, PU 및 PU-S상에서 혈장단백질의 응고시간 (PRT)을 조사한 결과, PU-S의 PRT는 PU 보다 길게 나타났다. 결과적으로 PU에 실리콘 세그멘트를 도입하므로서 항혈전성이 향상되었으겨 이것은 PU필름의 공기측면에 많이 노출되어 있는 soft 세그멘트의 역할에 기인한 것이라고 사료된다.
The polyurethane prepolymers, which were previously synthesized from 4,4'-diphenylmethane diisocyanate (MDI) and polytetramethylene glycol (PTMG), were chain extended by ethylene diamine or polydimethylsiloxane having amino group at both ends of the chain (AT-PDMS), giving polyetherurethaneurea (PU) and polyetherurethaneurea containing PDMS segment (PU-S), respectively. The interactions of synthetic polyetherurethaneurea with blood components were examined to compare in vitro blood compatibility of PU and PU-S. It was found from platelet adhesion study by lactate dehydrogenase that the amount of platelets adhered on the PU-S(22-24%) was smaller than that on the PU (32%). And the release of serotonin from platelets adhered was more suppressed on the PU-S than on the PU control. These results were well consistent with the results obtained by the observation of scanning electron microscope (SEM). Meanwhile, it was revealed from the experiment of plasma recalcification time (PRT) that PU-S showed a longer PRT than PU control. To conclude, by the introduction of polydimethylsiloxane segments to a PU, the antithrombogenicity of PU was improved and this might be attributed to the role of soft segments which largely exposed to air surface of PU-S film.
