γ-Secretase inhibitors which prevent Notch activation are emerging as potent therapeutics for various inflammatory diseases such as ischemic stroke and rheumatoid arthritis. However, their indiscriminate distribution in the body causes serious side effects after systemic administration, because Notch proteins are ubiquitous receptors that play an important role in cellular functions including differentiation, proliferation, and apoptosis. In this study, hyaluronic acid nanoparticles (HA-NPs) bearing a γ-secretase inhibitor (DAPT) were prepared as potential therapeutics for rheumatoid arthritis. From the in vivo biodistribution and therapeutic efficacy tests, we demonstrated that DAPT-HA-NPs have the good targetability to inflamed joints, resulting in high therapeutic efficacy. |
