제목 Angiongenic Effects of Demineralized Bone Matrix in the Repair of Calvarial Bone Defects
초록 This study investigated the influence of DBM on angiogenesis and osteogenesis in a calvarial defect model. DBM/PLGA scaffolds were manufactured by solvent casting/salt leaching method, and each scaffold contained 10, 20, 40, 80wt% DBM respectively. Bicortical holes were placed on calvaria of rats and then filled with scaffolds. Histologic analyses were carried out concerning number, cross-sectional area of vessels and area, density of regenerated bone. In all experimental groups, bone formation occurred in a typical centripetal direction and new vessel formation took place in front of the osteogenic regeneration front. The 20wt% DBM scaffold was superior its ability to regenerate new bone, induced more intensive angiogenesis and expressed a highly osteocalcin level. DBM not only attracts osteoblasts into the bony defect and stimulates their differentiation, but also leads to more intensive angiogenesis by means of angiogenic activation, which plays an important role of bone formation.
