Single yeast cell traps for monitoring chromosome dynamics |

Movement of chromosome conducts significant roles in various cellular process such as DNA replication, genetic recombination, epigenetic phenomena during mitosis and meiosis. Here, we present the microfluidic based single yeast cell traps for monitoring the movement of chromosome at meiosis (“prophase I”) stage. The existence of cell in the aperture trap makes the fluidic pressure difference which acts a driving force for sequential cell trapping. After trapping cells, sporulation medium is provided to induce meiosis at a specific time. In conclusion, we demonstrate the tracking and quantification of chromosome dynamics in yeast cells and obtain key measurement that the meiotic chromosome motion requires both actin and telomere/nuclear envelop association. We believe that this microfluidic approach will be useful for investigating other cell biological processes requiring single cell feature tracking and quantification. |
