The marine bacterium Saccharophagus degradans (Sde2-40) has an extraordinary ability to degrade various polysaccharides commonly found in the ocean, including cellulose. In this study, the growth and PHA production of Sde2-40 cells were observed in the aquarium salt medium supplemented with glucose or cellulose (avicel PH101) as a sole carbon source. PHA production by Sde2-40 was carried out in 5L fermenter using 10 g/L glucose or avicel as carbon source with 2L of sterile minimal medium. In fed-batch culture, a sterile solution of glucose or avicel was fed to recover the carbon source concentration. The maximum DCWs reached up to 4.20 g/L at 56 h for glucose and 2.11 g/L for avicel at 144 h. After the feeding, PHA concentrations gradually increased to 2.22 g/L at 72 h and 0.41 g/L at 144 h for glucose and avicel, respectively. Likewise, PHA contents for glucose and avicel in separate experiments were 52.8% and 19.2%, respectively. |
