Tuning of Coenzyme B12 Riboswitch sensor for signal amplification with toehold switch |

The riboswitches are attractivesensor modules for use in synthetic biology applications. However, the tuningof the riboswitches was constrained by their difficulties and narrow range ofmodulation. Therefore, novel molecular parts and strategies for straightforwardtuning of the fold-change of riboswitch circuits are needed. Here, we devised atoehold switch-based modulator approach that combines a hybrid input constructconsisting of a riboswitch and transcriptional repressor and de-novo-regulator,toehold switches. Optimization strategies that balancing the expression levelsof the toehold switch greatly improved the fold-change(261-fold). Furthercharacterizations confirmed low leakiness and high orthogonality of fivetoehold switch pairs, indicating the broad applicability of this strategy toriboswitch tuning. These results demonstrate that toehold switches can provideprogrammable and modular plug-and-play genetic parts for the response tuning ofriboswitch circuitry. |
