In a spore surface display system, the success of recombinant protein expression is highly dependent on the choice of anchoring motif. In this study, we developed an efficient Bacillus subtilis display system using novel anchoring motif derived from the coat protein CotY. And we inserted industrial enzyme α-amylase from B.subtilis at the C-terminal end of anchoring motif. We checked the surface expression of α-amylase using flow cytometry. Histogram of flow cytometry showed higher fluorescence intensity, possibly due to their outer location on Bacillus spore. We measured whole cell enzyme activity of wild type (DB104) and recombinant cell (DB104-CotY-AmyE). The enzymatic activity of the spore expressing α-amylase was more than 10 times higher than that of the host spore. |
