제목 Preparation of microfluidicsorter for screening of Yarrowia Lipoltica producing high lipid accumulation
초록 Asbiofuels, they are a perfect substitute to liquid fossil fuels with respect tocost, renewability, and environmental concerns. Biofuels derived frommicroalgae showed promising potential as a future renewable bioenergy source. Here,we demonstrate a screening randomly mutated Yarroiwa Lipolytica library and sort outthe target subsets exhibiting higher growthrate and increased lipid production by using valve based microfluidic sorter (μFADS). First,we generate the cell-encapsulated alginate droplets through a flow-focusing deviceand perform the gelation process through calcium ion releasing by lowering pHcondition. Then, the Y. Lipolyticacells were cultured in alginate gels that functioned as an individualmicro-reactor and re-injected into μFADS to evaluate their capacity and sort target cells. As a result, the alginate gelscontaining cells showing 1.5 – 2 times higher growth and lipid production ratewere selectively retrieved compared to the cells in the waste channel.
