제목 Hydrogel as a matrix to support isolation, propagation, differentiation and transplantation of adult stem cells
초록 Adipose tissue (AT) represents an abundant, practical source of donor tissue for stem cells, so it has been called as “Gold Mine” of stem cells. The conventional method to isolate AT-derived stem cells (ASCs) is monolayer culturing of the stromal vascular cells after collagenase digestion. This enzymatic method results in significant variation of yields and also requires additional steps to remove unwanted cells and debris. In this study we developed 3-dimensional (3D) organ culture to isolate and expand of ASCs without tissue dissociation step. The AT was completely embedded in fibrin or collagen gel. The cells which were migrated and proliferated from AT into the hydrogel could be successfully isolated by substrate-specific protease treatment. The yield of cell isolation by our novel method was approximately 100-fold higher than enzymatic method and cells retained adipogenic, osteogenic, chondrogenic, musculogenic, neurogenic and endothelial differentiation potentials.
