Toxicity Evaluation of Polystrene Microplastics Using Human Fibroblast-Derived Spheroids |

Owing to severe environmental accumulation of microplastics (MPs), many efforts have been made to investigate potential toxicity of the MPs using cells and model animals. Although many studies have shown physical and chemical toxicities of MPs in cellular level, most of researchers have utilized using a 2D cell culture model, which may differ from a real in-vivo animal model. Herein, we investigate the cellular level toxicity of polystyrene (PS) microplastics with 3D cultured human dermal fibroblast (HDF) spheroids. Diverse sizes from nano to micrometers of the MPs are exposed to the HDF spheroids and their penetration into the spheroids are characterized by confocal live cell imaging. Then, morphological changes in the spheroids and the related biomarkers are evaluated to compare with 2D cell culture models. We expect that this approach provides a new perspective on the toxicity of MPs under the conditions closer to in-vivo. |
