We present aPEptide-Directed Photocrosslinking (PEDIP) method for the site-specificconjugation of IgG to protein toxins to generate antibody–protein toxinconjugates (i.e., immunotoxins). To develop the PEDIP reaction, we focused onthe Fc-specific binding peptide FcIII and a photoreactive amino acid analogue,p-benzoylphenylalanine (Bpa), which was used to replace one of the positions ofFcIII to generate covalent bonds to the Fc region of the antibody. Site-specificallyconjugated homogenous immunotoxin products could be generated, regardlessof the type of antibody, by the PEDIP method. The therapeutic efficacy andtoxicity of immunotoxins generated by this method were confirmed by in vitrocell-based assays and a mouse model. |
