Development of efficient gene delivery vehicles using Polyethylenimine and Mannitol copolymer, improving transfection efficiency and cytotoxicity. |
초록 |
Polyethylenimine (PEI) has been used as a golden standard among non-viral gene delivery systems. Recently, modification of PEI has been an important issue to target not only cell type-selective gene transfer but also intracellular compartment-selective transfer because the way of PEI to enter the cell markedly influences its gene delivery efficiency. We have designed and synthesized polymannitol-based transporter (PMT) to have lower cytotoxicity by degradable ester group and higher transfection efficiency by osmolyte polymannitol. We utilized mechanosensing signal transduction of the cells to shift the uptake route into the caveolae-mediated endocytosis. Caveolae-mediated endocytosis was reported to lead internalized molecules to less destructive pathway which makes it more advantageous way to deliver gene than clathrin-mediated endocytosis. By controlling endocytosis route, we will expect to regulate the intracellular fate of the PMT for higher gene delivery efficiency. |
