Injectable system is one of the very attractive fields in drug delivery. This system has useful advantages such as ease to application, localized delivery for site specific action, minimized side effects and patient compliance. Chitosan based thermosensitive hydrogel as an injectable drug carrier was prepared by grafting N-isopropylacrylamide(NIPAAm) onto chitosan backbone. This copolymer possess not only biocompatibility and biodegradability as useful properties of chitosan but also LCST around 32℃ originated in thermosensitivity of PNIPAAm. In addition, drug loaded biodegradable PLGA microparticles were prepared by the double emulsion method suitable for incorporation of hydrophilic drugs. Thermosensitive hydrogel can be incorporate easily various types of drug as an injectable carrier through sol-gel transition behavior. However, there is a problem that the suitable control of the release kinetic for the therapeutic effect is difficult in the case of low MW drugs. In this study, the double barrier system using thermosensitive chitosan and biodegradable PLGA microparticles was developed. 5-fluorouracil and bovine serum albumin as model drugs were used and their release behaviors were investigated.
