Controlled Release of Dexamethasone from Closed Biodegradable PLGA Microspheres |

Microspheres have been utilized as a drug delivery system in the biomedical applications that need constant drug concentration in the blood or promote differentiation of specific cells. Poly (lactic-co-glycolic acid) (PLGA) copolymer is a synthetic biodegradable polymer which can encapsulate both hydrophilic and lipophilic drugs. Dexamethasone (Dex) has been shown to be essential for chondrogenic differentiation of stem cells. In this study, PLGA microspheres were fabricated by W/O/W method using H2O2 as a gas foaming agent. Oxygen gas bubbles was generated with the aid of catalase, a H2O2 decomposition catalyst. This method is simple and environment-friendly, furthermore it is expected sterilizing effect. Various conditions were examined in the preparation of microspheres and the resultant specimens were characterized. Dex was loaded during the fabrication of microspheres and the release pattern of dex was examined in different surface morphology of PLGA microspheres. |
