Evaluation of different osteogenic potentials of naturally-derived extracellular matrices (ECM) in 2D configuration |
초록 |
Extracellular matrix (ECM) is a physical entity that is a complex and highly ordered array of cell-derived proteins and other biomolecules. Its hierarchical structure provides a natural environment in which cells can survive and function properly. The primary focus is to prepare cell-derived matrices (CDMs) from different cell sources and to assess their effects as a physical cue in controlling the osteogenic activity of preosteoblasts. Our hypothesis is that each CDM may play different roles in eliciting cellular responses because cells would create individually unique microenvironment. The CDMs showed that cells could secrete the matrix components and utilize them to build a naturally assembled architecture. Interestingly, fibronectin was assembled in much different way as compared to type I collagen. This study strongly hinted that each CDM had different osteogenic potentials as supported by the results of histology, ALP activity, and gene expression. |
