Microalgaeare attracting attention as a bio-platform for biological conversion ofindustrial CO2 based on its excellent carbon fixation ability.However, due to the low productivity of sunlight-driven cultivation system, commercializationof microalgae process is challenging. Light fluctuations are considered to bethe major causes of cell growth reduction. Here, a novel microdroplet screeningsystem capable of effectively delivering external light conditions toindividual cells was introduced to select mutants exerting optimal growthperformance under fluctuating light conditions. After cultivation of randominsertional mutants in the gel microdroplet photobioreactor, high-density dropletswere selected via density-gradient centrifugation. Selected mutant strainshowed improved cell growth and photosynthetic efficiency in fluctuating light environments.For proof-of-concept field culture, the superior mutant strain was applied inoutdoor mass cultivation using flue gas CO2. |
