제목 PLGA Microspheres Incorporated with TGF-β1/Chondroitin Sulfate Ionic Complex for Cartilage Tissue Engineering
초록 Transforming growth factor-β (TGF-β) is a cytokine, which has an autocrine and paracrine action in the testis. It also displays potent immunoregulatory and anti-inflammatory activities. TGF-β1 with an isoelectric point (IEP) of 9.5 is an isoform of TGF-β that plays an important role in the growth and differentiation of articular cartilage. Chondroitin sulfate (CS) is a member of the glycosaminoglycal (GAG), an acidic mucopolysaccharide found in cartilage, skin, corneas, and umbilical cords. TGF-β1 is positively charged and forms a polyionic complex with negatively charged acidic CS (IEP of 5.0) at pH 7.4. In this study, the ionic complex of TGF-β1/CS was formed with different fractions of CS and TGF-β1. Poly(lactide-co-glycolide) (PLGA) microspheres incorporated with TGF-β1/CS ionic complex were fabricated by emulsion-solvent evaporation method. Various conditions were tested and the resultant PLGA microspheres with the ionic complex were characterized.
