Bioreducible Crosslinked Hyaluronic Acid Nanoparticles for Targeted Cancer Therapy and Imaging |

Self-assembled polymeric nanoparticles with high in vivo stability and tumor-targeting ability have been developed as drug carriers for improved therapeutic efficacy. In this study, we developed bioreducible core-crosslinked hyaluronic acid nanoparticles (HANPs) by a facile method in aqueous conditions. The resulting nanoparticles (CC-HANPs) were smaller in size than HANPs owing to the formation of a compact core-cross-link. The CC-HANPs exhibited enhanced structural stability under diluted condition with PBS containing FBS and sodium dodecyl sulfates (SDS). In particular, the drug release rate of CC-HANPs was rapidly accelerated in the presence of 10mM GSH, whereas the drug release was significantly retarded in physiological buffer (pH 7.4). Overall, these results suggest that bioreducible CC-HANPs could be applied as a targeted doxorubicin delivery carrier for cancer therapy and imaging. |
