1,1-Difluoro-1,3-enyne derivatives are important building blocks for the synthesis of fluorinated compounds having unique biological and physical properties. 2-(t-Butyl)dimethylsilyl-1,1-difluoro-2-iodoethene 1, a precursor of the title compound, was prepared in high yield from the reaction of 2,2,2-trifluoroethyl iodide with 2 equiv of LDA at low temperature, followed by treatment with (t-butyl)dimethylsilyl chloride. Further Stille cross-coupling reaction of 1 with various alkynylstannanes in the presence of Pd(PPh3)4 and CuI afforded the corresponding 2-(t-butyl)dimethylsilyl-1,1-difluoro-1,3-enynes 2 in good yields. Further acylation reaction of 2 with acyl chloides in the presence of AlCl3 will be presented. |
