Phenylpropanoids are derived from aromatic amino acids and medicinally important plant natural products. In this study, we report the bioproduction of two representative phenylpropanoids, coniferyl alcohol (CA) and dihydroquercetin (DHQ) by engineered Escherichia coli. First, by employing hpaBC , OMT1, 4CL4 and CCR1, CA producing strain produced 187.7 mg/L of CAs. Next, 239.4 mg/L of DHQ was produced by introducing F3H, TT7 and CPR. Signal peptide of cytochrome P450 TT7 was manipulated for higher production. In addition, to produce flavonolignans from glycerol, ascorbate peroxidase was introduced to produce 0.04 μg/L and 1.29 μg/L of silybin and isosilybin respectively by stepwise culture. Finally, 0.12 μg/L isosilybin was produced directly from glycerol, with a single strain expressing 16 essential genes.
