Improvement on Solubility and Dissolution rate of Rivaroxaban by Supersaturatable Self-microemulsifying Drug Delivery System |

Rivaroxaban is a potent selective oral direct factor Xa inhibitor, which has been shown poorly aqueous solubility and low bioavailability. The purpose of this study was to develop supersaturatable self-microemulsifying drug delivery system (S-SMEDDS) for improving solubility and dissolution rate of rivaroxaban. An efficient self-microemulsifying carrier for rivaroxaban was selected and optimized using solubility testing and phase diagram construction. The formulations were characterized by assessing self-emulsification performance, droplet size, in vitro drug release and stability. Rivaroxaban has been shown high solubility in Labrafill m 2125, Tween 80 and tetraglycol. Overall, incorporation of hydrophilic polymer(Soluplus, PVP K 30 and Solutol HS 15) in SMEDDS resulted in improved solubility and dissolution rate compared to pure rivaroxaban. This study suggests that S-SMEDDS may be promising for enhancement on solubility and dissolution rate of rivaroxaban in the future development. |
