Osteoinductive Hyaluronic Acid-Catechol Hydrogel to Enhance Bone Regeneration by Human Adipose-derived Stem Cells |

Development of osteoinductive biomaterial scaffolds is critical for efficient bone formation following stem cell transplantation. Here, we report a catechol-functionalized hyaluronic acid (HA-catechol) hydrogel with improved biocompatibility compared to typical photopolymerized HA hydrogel. To improve osteoinductive property of HA-catechol hydrogel, the peptides derived from BMP-2 were incorporated into the HA-catechol hydrogel. BMP-2-incorporated HA-catechol hydrogel greatly enhanced in vitro osteogenic differentiation of human adipose-derived stem cells (hADSCs) in both osteogenic and nonosteogenic medium conditions. Finally, transplantation of hADSCs using BMP-2-HA-catechol hydrogel enhanced in vivo bone formation in a critical-sized calvarial bone defect.
