Growth Factor-immobilized Biodegradable Polymer Scaffolds for Cartilage Regeneration |

Polymer scaffolds with controlled release of growth factors may be effective for tissue regeneration. It is recognized that many growth factors and cytokines possess affinity for heparin. Heparin is recognized to have specific affinity with various polymers. In this study, polycaprolactone (PCL) scaffolds were fabricated by blending PCL with a small amount of Pluronic F127 by a melt-molding particulate-leaching method. The Pluronic F127 chains exposed onto the scaffold pore surfaces was used to bind heparin and the following growth factors (TGF-β2, BMP-7). The TGF-β2 and BMP-7 were shown to be effective for the differentiation of bone marrow- or fat-derived mesenchymal stem cells to chondrocytes and thus cartilage regeneration. Amounts of heparin and growth factor in the PCL/F127 scaffolds were quantified by Toluidine blue method using UV spectroscopy and by ELISA kit, respectively. The growth factor release behaviors from the growth factor-immobilized scaffolds were investigated. |
