Preparation and Characterization of Plasmid DNA-LoadedPLGA/Pluronic F127 Scaffold |

In recent, the cartilage regeneration using tissue engineering technique and local gene delivery system have been considered as one of the promising therapeutic methods. In this study, we prepared plasmid DNA-loaded PLGA scaffold to enhance the chondrogenic differentiation of mesenchymal stem cells (MSCs). For more effective plasmid DNA release from the scaffold and the following transfection into cells of plasmid DNA, the PLGA scaffold was hydrophilized by Pluronic F127 (can allow faster release behavior of plasmid DNA than hydrophobic one) and the plasmid DNA was complexed with polyethyleneimine-polyethylene glycol (PEI-PEG, non-viral vector). The release behavior of the plasmid DNA/PEI-PEG complex from the hydrophilized PLGA/Pluronic F127 scaffold, transfection efficiency of the plasmid DNA/PEI-PEG complex into MSCs, and the cytotoxicity of the cells were investigated.
