Preparation of nano-emulsified paclitaxel for sustained release. |
초록 |
Purpose. To prepare nano-emulsion of paclitaxel by self micro emulsion drug delivery system(SMEDDS) and confirm the stability of nano-emulsified paclitaxel according to MPEG-PLGA diblock copolymers. Methods. Lactide and glicolide were added to MPEG dissolved in toluene under nitrogen, followed by the addition of stannous octoate in dried toluene. The stability of nano-emulsified paclitaxel was examined by shaking. Results. The polymerization gave MPEG-PCL diblock copolymers with the various ratios in MPEG and PLGA. Paclitaxel and MPGE-PLGA diblock copolymers were dissolved by solubilizers which were a mixture of tetraglycol, Cremophor ELP, and Labrasol. The droplet size distribution is comparatively narrow for all formulations. This indicated that stability of nano-emulsion strongly depended on the HLB values of MPEG-PLGA diblock copolymers. Conclusion. These results prove that the formulations tried in this study could be used as administration form of paclitaxel for clinical trials. |
