To develop reactive DNA oligomer, wefocused on use ofoxanine (Oxa),one of mutagenic lesions, whichis produced fromnitrosative deaminaiton of guanine. SinceOxa hasa kind of activated carboxyl group (O-acylisourea formation) in the base ring,Oxashowsamino group-directed reactivity, whichisusefulforlinkage of DNA probe on the surface.We set upthe synthesis procedurefor chemical preparation of Oxa-containing DNA oligomer (Oxa-DNA)and fabricated DNA microarray by ink-jet spotting of this Oxa-DNA probe on NH2-functionalized glass-slide.Reactive DNA oligomerand its immobilized system willbe usefulfordevelopingadvanced nano/biotech systems. |
