Generally, it is regarded that hydrophilic macromolecules such as proteins, peptides, and vaccines have been limited to penetrate across skin. In this study, we applied chitosan-conjugated, Pluronic-based nano-carriers as an efficient delivery vehicle of hydrophilic proteins across human skin. The in-vitro permeation experiments through human cadaver skin revealed enhanced permeability of hydrophilic proteins such as FITC-BSA and FITC-Insulin by direct penetration of the nano-carrier across skin. In addition, the bioactivity post-permeation of proteins via the functional nano-carrier was also confirmed by delivering ß-galactosidase, and maintained well.
