제목 | Application of cellobiose dehydrogenase from Phanerochaete chrysosporium in whey enzymatic fuel cell |
초록 | Cellobiose dehydrogenase (CDH), as an extracellular enzyme production of fungi, has been reported and applied for degradation of lignocellulose. It has a unique structure consists of two distinct domains, flavin domain (DH) and haem domain (CYT) which is connected through interdomain electron transfer (IET). Based on this unique enzyme structure, CDH has been received high interests in electrochemical applications such as enzymatic fuel cells (EFC) and biosensors. In this study, heterologous expression of the cdh from P. chrysosporium KCCM 60256 in Pichia pastoris was achieved for efficient production of the enzyme. To achieve an EFC system, graphite oxides were immobilized on the Au surface of both electrodes; recombinant CDH (PcCDH) and laccase were modified on anode and cathode, respectively. The reaction conditions of the EFC were 50mM whey at pH 5.0 in 0.1 M potassium phosphate buffer. The potential prospect of whey and CDH application in EFC system has been successfully demonstrated by this study. |
