제목 Evaluation of various types of scaffold for tissue engineered intervertebral disc using cultured annulus fibrosus
초록 We are trying an alternative approach that consists of a functional IVD composed of annulus fibrosus seeded to various scaffolds using tissue-engineering principles. DBP/PLGA, SIS/PLGA, SIS/DBP/PLGA and PLGA scaffolds were prepared by solvent casting/salt leaching. SIS sponge was fabricated by freeze-drying . AF cells were seeded into scaffolds with 2x105 cells. It was evaluated cell proliferation by MTT assay. The implants were removed after 1, 4 ,6 weeks and observed their gross morphology Histological section were confirmed H&E, Safranin-O, Masson's trichrome staining. We observed specific gene expression of disc cell by RT-PCR. Proliferation of AF cells were observed that the PLGA/SIS/DBP scaffold high. Scaffolds maintained theirs morphology. It could confirm aggrecan, type∏ collagen expression by RT-PCR.
