Composite Microwell Arrays via Controlled Photo-polymerization in Microfluidic Device |

Hydrogel-based microfluidic platform has been extensively investigated because it is easy to position at microfluidic system by photo-polymerization and can perform both sensing and actuation functions. Here we propose novel method for preparation of composite microwell arrays with response to external stimulation via in-situ polymerization in microfluidic device. Microwell array with two different polymers was fabricated using two step sequential photo-polymerization; polymer with no responsive characteristics as a supporting layer and pH-responsive hydrogel as an actuation layer. Without using photo mask which needs complicated mask aligning process, silicon microdot array was used to define the structure of two polymer layers. Composite microwell array was actuated with pH buffer solution to give complete entrapment of analytes in each well. Hydrogel microwell demonstrated in this study is expected to be utilized in multiplex biomolecular analysis. |
