In the treatment of cardiovascular disease through artificial blood vessels, even small amounts of blood clots can block the vessels. As a countermeasure for this, we used a strategy to connect the cells to the surface of the vessels by attaching the molecules that binds specifically to endothelial progenitor cell(EPC). That molecule is a peptide with an sequence of TPSLEQRTVYAK(called TPS peptide). We choose the poly(lactide-co-ε-caprolactone) as material and covalently immobilized that with TPS peptide for recruiting EPC and differentiating to endothelial cells. In addition, because when plasma proteins attach to peptide, the binding ability is deteriorated, we also connect the heparin with peptide to the polymer. That graft were implanted subcutaneously in the rat and were operated surgery in the rat aorta. In experiment result, we can link the TPS peptide and heparin to the PLCL polymer by covalent bonding, we find that which can be an effective strategy for preventing thrombosis. |
