In the treatment of cardiovascular disease through artificial blood vessels with small diameter(<6mm), even small amounts of blood clots can block the vessels. As a countermeasure for this, we used a strategy to derive the cells to the surface of the vessels in situ, by attaching the molecules that binds specifically to endothelial progenitor cell(EPC). That molecule is a peptide with an sequence of TPSLEQRTVYAK(TPS peptide). And we choose graft material as a ‘star-shape poly(L-lactide-co-ε-caprolactone)’ for using many functional group to connect TPS peptide. In addition, because TPS's binding ability with EPC is deteriorated when blood plasma proteins attach and block the peptide, we also connect the anticoagulant pharmacy: heparin to the polymer with TPS. In conclusion, in situ surgical experiment showed that many EPC cells became EC cells and greatly prevented thrombosis. |
