제목 Physical Properties of Collagen Gel Solutions and Collagen Matrices using SIS Powder for Cell Culture
초록 Small intestinal submucosa (SIS) is an unusual tissue that a naturally occuring extracellular matrix rich in collagen has been successfully used as a soft tissue graft in several body organ systems. SIS was isolated from porcin intesine and lyophilized. Lyophilized SIS was powdered by freezer-mill. After SIS powder dissoved in 0.5M acetic acid, SIS solution were treated with different concentrations (0-0.2%) of glutaraldehyde for 24h. The viscoelasticity of the resulting collagen gel solution was measured using dynamic mechanical analysis (DMA) and determined degree of crosslinking of collagen with different concentrations of glutaraldehyde using 2,4,6-trinitribenzenesulfonic acid. These crosslinked collagen gel solutions were freeze-dried to for a matrix. The break modulus of these collagen matrices measured by DMA and morphology of them observed by scanning electron microscope (SEM). The cell compatibility of NIH3T3 with these collagen matrices was found to be acceptable at a cell density of 2×105cells/㎠.
