제목 | Evaluation of cells on polymer surface with different wettability by RT-PCR analysis |
초록 | The adhesion and proliferation of mammalian cells on polymeric biomaterials depends mainly on the surface characteristics such as wettability, chemistry, charge, roughness, and rigidity. A large number of research groups have studied the interaction of cultured cell and various polymers with different wettabilities to correlate the relationship between surface wettability. In this study, we have recognized that adhesion and proliferation of cells on polyethylene (PE) films depended on the surface properties mainly focused on analyzing the RT-PCR pattern of expression exhibited by several growth-associated genes. Plasma treatment of PE films was developed to increase surfaces wettability and to improve adhesion and proliferation of NIH/3T3 fibroblasts. The water contact angle of the film surfaces gradually decreased with increase of plasma treatment time. The surface-modified PE films were characterized by the measurement of the water contact angle and ESCA. The adhesion and proliferation of cells on PE films with different wettability were characterized by SEM, cell count, and RT-PCR. Analyzing the pattern of expression was compared with several growth-associated genes (e.g., c-fos, c-myc, and p53) in NIH/3T3 fibroblasts onto PE surfaces with different wettability.
