제목 Chondrogenesis of Human Adipose-derived Stem Cell using Pluronic F127-HA composite Hydrogel
초록 Composite of Pluronic F127 and hyaluronic acid (HA) was investigated, exploring the benefits of each hydrogel in the chondrogenesis of human adipose-derived stem cell (ASC). F127 was chemically modified through a series of reactions to graft heparin and then dexamethasone (Dex), respectively. Crosslinked HA was used as a physical stabilizer of F127. The composite F127/HA was physically coupled to TGF-1. We hypothesize that as the physical stability of hydrogels is maintained, chondrogenesis can occur with the aid of the incorporated chondrogenic factors. DAPI staining showed that ASCs were widely dispersed inside the hydrogel matrix. Immunofluorescent staining of type II collagen indicated that the group, TGF/Dex conjugated F127/HA sustain more cartilage-specific protein than F127/HA. This system proved that the incorporation of chondrogenic inducers was effective in inducing chondrogenic differentiation of ASC.
