Characterization of Purified Alginate Sponge for Three-dimensional Culture Effect |

Alginate is a natural polymer that is frequently used in biomedical applications. But alginate is caused immune rejection in the body. To overcome this problem, we have developed using modifed Korbutt’s method for alginate purification. NIH/3T3 and RAW 264.7 macrophage cells were seeded in purified and non-purified alginate sponges. Cell viability analyzed by MTT assay after 1, 3 and 5 days. Cell proliferation and cell morphology analyzed by SEM assay after 1, 3 and 5 days. The Mechanical properties of alginate sponge was measured by Compressive strength. The MTT result of purified alginate sponges showed that the cellular viability was excellent. The SEM result of purified alginate sponges showed that the cell proliferation and cell morphology was excellent. This result indicates that purified alginate sponge might be effective for cell environment.
