Tissue engineering has the possible to improve upon current techniques for intervertebral disc repair. In this study, purified alginate sponge were used in intervertebral disc regeneration. Annulus fibrosus(AF) cell were seeded in purified alginate(PA) and non-purified alginate(NA) sponge, and then cell viability analyzed by MTT assay. RT-PCR was assessed to measure mRNA expression for type I, II collagen, aggrecan from AF cells in PA and NA sponge. AF cell-loaded PA and NA sponge were implanted into the back of BALB/c-nu nude mouse, and the implanted alginate sponge were extracted after 4 weeks. The alginate sponge evaluated by histology through H&E, type I, II collagen, Safranin-O staining. When we cultured AF cell of rabbit, we observed better to keep the characteristic of AF cell in the PA sponge than NA sponge. This result indicates that PA sponge is useful for intervertebral disc regeneration. This study is supported by WCU(R31-20029) and MBC (0405-BO01-0204-0006). |
