We demonstrate the droplet based microfluidic platform for analysis of antimicrobial susceptibility at single cell level. First, we make the single cell encapsulated microdroplet generator integrated with linear concentration gradient to control the composition of antibiotic mixture. Using this strategy, we tested gentamicin against gram negative bacteria (i.e. E.coli O157:H7 ) to determine the minimum inhibitory concentration(MIC). Screening the growth profile of cells in droplets having different concentration of gentamicin resulted in MIC determination in only 4-6 h using our device. Additionally, we compare the minimum inhibitory concentration result from bulk system using an Erlenmeyer flask. This result show that our platform has the great potential to be used for antimicrobial susceptibility testing (AST) with other bacteria (i.e. gram-positive bacteria) and antibiotics. |
