Effect of DBP in DBP/PLGA scaffold for Articular Cartilage tissue engineering |

A solvent casting/salt-leaching method for the preparation of DBP/PLGA scaffolds was designed by varying the DBP contents with 0, 10, 20, 40 and 80 wt %. The DBP contents of the DBP/PLGA scaffold influenced the cell viability and extracellular matix secretion. Artiular chondrocytes of Newzealand White Rabbits were in vitro cultured for 3 weeks. To charterize chondrocyte of MTT, GAG assay, RT-PCR, H&E, Safranin-O stanining were evaluated. As DBP contents of these scaffolds become higher, the rate of cell growth and the amount of GAG increase. The DBP/PLGA scaffolds with DBP content from 10 to 20 wt % are better cell proliferation, ECM secrection and the gene expressions of chonrocyte. Chondrocytes prefer the group of PLGA scaffolds with DBP contents from 10 to 20 wt % for better proliferation and ECM production. It is believed that DBP would significantly affect the cell metabolism. This study is supported by WCU(R31-20029) and MBC (0405-BO01-0204-0006). |
