Hemostatic activity of thrombin loaded plant derived oxidized nanocellulose-gelatin based scaffolds |

During different surgery, transplantation or post-surgical hemorrhage and severe bleeding in traffic accident are the main reason for trauma death. Excessive bleeding control is much important in surgery or any other trauma injury. In this work, TEMPO-oxidized nanocellulose (TOCN)/ gelatin (G) composite sponge was successfully prepared in co-operation with thrombin (Th) and their use as hemostatic material was investigated. Fourier transform infrared (FTIR) spectra confirms the presence of carboxyl group on TOCN surface and incorporation of gelatin and thrombin in the composite. Physical characteristics of the composites were observed where uniformity, porosity, water absorption ability, swelling rate, degradation behavior had relationship with the percent gelatin incorporation. Among the samples TOCN 2.5G-Th composite sponge had excellent hemostatic effect, lowest hemostatic time and lower blood loss in liver avulsion model. In addition, hemocompatibility and cytocompatibility of these sponges were also evaluated where addition of thrombin did not hamper any of the assay. In fact, thrombin enhances blood coagulation and blood cell aggregation with the prepared sponges compared with other non-thrombin loaded hemostatic sponges. |
