Multi-stimuli Responsive Elastin-Calmodulin Diblock Polypeptides for Dynamic Drug Delivery Systems |

Protein block copolymer-based nanostructures have gained increased interest for a variety of biomedical applications because of easy control over amino acid sequence and block length and facile introduction of specific biological domains with stimuli-responsiveness at genetic level. We report herein molecular design, expression and purification of multi-responsive elastin-based polypeptide (EBP)-calmodulin (CalM) diblock polypeptides and characterization of thermal- and ligand responsiveness of the EBP and CalM block. Temperature-triggered self-assembled micelles of the EBP-CalM diblock polypeptides were formed via physical crosslinking due to thermal responsiveness of EBPs, and their micellar structures were affected by calcium and ligand. These multi-stimuli responsive EBP-CalM diblock polypeptides which would allow stimuli-triggered drug release kinetics are useful for dynamic drug delivery systems. |
