Recently, stem cellspheroid have attractive compare to two-dimensional (2D) cell culture. However,spheroid differentiation is difficult to form since the cell capability fornutrient uptake would be altered. Therefore, the stem cell spheroid (SCS)density should be managed to establish its differentiation. In here, we reporta 3D monitoring by chemical analysis of odontogenesis of hDPSCs spheroids basedon Raman spectroscopy. Increase of cell density reduced the growth mediumdiffusion as proven by the decrease of Raman signal. Also, the nutrientdistribution upon odontogenesis was investigated by performing Raman mappinganalysis to reveal the correlation between cell density and the successfulratio of differentiation. Hence, Raman analysis is advantageous for analyzing3D cell culture, which will be useful for drug discovery. |
