Stem cell spheroid cultures using conventional methods including spinner flask or low attachment plate have limitations that mass production and size control of spheroids are impossible. Therefore, many studies tried to overcome those limitations by using patterned hydrogels which are composed of agarose, polyethylene glycol, etc. However, it has a drawback that accessibility is inferior due to their complex platform construction process. Thus, we developed hydrogel embossing technique for easy fabrication and culture of human adipose derived stem cell (hADSC) spheroids by using commercially available sandpaper. It was confirmed that irregular surface protrusions of hydrogel keep the narrow size distribution of the hADSC spheroids. Furthermore, we could utilize the hADSC spheroids from hydrogels with embossed surface for 3D bioprinting. Our results showed that hydrogel embossing technique can be an effective tool for stem cell spheroid culture. |
