We proposed polyethylene glycol (PEG) coated mesoporous silica as mucoadhesive carriers for sustained delivery of dexamethasone (DM). For this, the SBA-15 microparticles were synthesized using pluronic P123 and cetyltrimethylammonium bromide as a surfactant. To increase the loading amount of DM, SBA-15 were functionalized with amine groups via the post grafting method, using 3-aminopropyl triethoxysilane. Then, we encapsulated DM into the amine-functionalized SBA-15 and coated DM-SBA-15 with PEG to allow a mucoadhesive property. As we evaluated the in vitro drug release profile in pH 7.4 PBS, the particles slowly released the drug for 24 h after a burst release of 45% on the first 20 min. To examine the mucoadhesive property, 4 mg particles were suspended in the 2 ml DI (1 mg/mL) containing type Ⅱ mucin for 0.5, 1, 24 h, which revealed that twice much mucin was adsorbed to the PEG coated silica particles as compared with the noncoated particles. |
