Many of the biologically active secondary metabolites contain deoxysugars as structural components, and often these deoxysugar components are essential for the biological activity of the parent natural products. TDP-4-keto-6-deoxy-D-glucose is produced from thymidine-monophosphate with glucose-1-phosphate in large quantity by one-pot reaction. The reaction is required thymidine monophosphate kinase, acetate kinase, TDP-glucose synthase and TDP-D-glucose-4,6-dehydratase. Various TDP-deoxysugar (TDP-L-rhamnose, TDP-4-keto-2,6-deoxyglucose and TDP-olivose, …) moieties were synthesized from TDP-4-keto-6-deoxy-D-glucose with TDP-glucose 4-ketoreductase, TDP-glucose 3,5-epimerase, TDP-glucose 3-ketoreductase, TDP-glucose aminotransferase and TDP-glucose 2,3-dehydratase in vitro. Novel antibiotics are synthesized from pikramycin and methylmaycin with TDP-deoxysugar derivatives in vivo. |
