Cell-Laden Alginate Microgels Created by Controlled Coalescence of Dual-Cored Double Emulsions |

Alginate micrgels represent one of the promising carriers for cell encapsulation. These carriers are required to have controlled size and shape for successful transplantation of functional cells. Although conventional dropping method can produce alginate beads, it only achieves limited control over size. To overcome this limitations, we design a novel microfluidics platform which is able to create non-spherical double emulsion with two distinct dual cores. One of the core contains sodium alginate and the other contains calcium ions. Two cores enclosed by ultra-small volume of oil shell are brought into a contact by strong capillary force exerted by non-spherical envelop and eventually coalesce to form single-cored double-emulsion drops. This leads to ionic gelation within the single core to form calcium-alginate microgel. The microgels are rapidly dispersed into continuous water phase by rupture the oil shell, which provides high cell viability by minimizing contact with oil. |
